    Antiserotonin-antihistamine agents in allergic diseases clinical evaluation of cyproheptadine

    Recently a Chemical compound, l-methyl-4-(5-dibenzo-[a, e] cycloheptatrieny- lidene)-piperidine (cyproheptadine), was synthesized which presenta the characteristic of being at the same time a potent antihistamine and an antiserotonin substance with similar activity to that of lysergic acid diethylamide (Stone et al. [1]). It is well known that histamine plays an important role in the phenomena of allergy and anaphylaxis. However, as Humphrey and Jaques (2), Rocha e Silva (3) and numerous authors have shown (4,5,6,7), especially in anaphylactic reactions several other pharmacodynamically active substances in addition to histamine, such as 5-hydroxy- tryptamine or serotonin, bradykinin, etc., are released. Although the role that serotonin plays in allergic reactions is not clear, nevertheless it has been of considerable interest to investígate clinically a drug that has combined antihistamine and antiserotinin activity. The purpose of the present investigation was to study the effect of cyproheptadine on two types of allergic diseases: asthma, in which antíhistaminic agents have little favorable effect, and urticaria, in which the antihistamines exert considerable therapeutic effect.

    Epidemic hecatomb in the new world

    The American population developed, during thousands of years, free of epidemics that had been attacking Europe, Asia and Africa. The European and African migrations, after Columbus's first trip, produced an epidemic invasion of influenza, smallpox, measles, yellow fever, malaria, diphtheria, typhus, and other diseases that attacked the immunologically virgin populations and produced a very high mortality, with a diminution of the indigenous population of more than 90% in many places. According to historical evidence, the first epidemic was influenza, produced by swine strain of virus, immediately followed by smallpox. The Spaniards mated freely with the Indians producing a mixed race called the Mestizo, who were immunologically more capable of defending themselves against various viruses, bacteria, and parasites brought over from the Old World. Marriage between the races also was sanctioned by Queen Isabella (1503) and Fernando I (1515). With these new genetic immunologic defenses against infections, the Mestizo eventually made up the majority of the population of Indians in the New World.

    Erworbene Antigentoleranz und Anaphylaxie

    Medawar (1—4) und Burnet (5-9) haben neben anderen Autoren auf experimentel- ler Basis den Begriff der „inmunitaren Toleranz" entwickelt; auBerdem hat Burnet eine neue Inmunitatstheorie formuliert, die er Klonal-Selektion nennt. Wenn man einem fetalen oder neugeborenen Tier Zellen eines anderen Tiers glei- chen Zustandes von einem Zelltyp, der direkte Beziehung zur Antikórperproduktion hat, etwa Lymphozyten oder Suspensionen von Milzgewebe, einspritzt, entwickelt sich nach den Angaben obiger Autoren in einem solchen Tier ein Zustand von „inmuni- tarer Resistenz". Dieser erlaubt ihm dann Transplantate von Haut oder anderen Ge- weben des Tiers anzunehmen, das die Lymphozyten oder Milzzellen gespendet hatte. Nach Burnets Theorie (8-10) kónnen Tiere im Embryonalzustand Zellen oder Gewebe eines andern Tiers aufnehmen ohne Antikórper zu bilden, die diese zerstóren würden, wahrend dies im erwachsenen Zustand der Fall ist, wo Heterotransplantate nicht an- gehen. Wenn aber der embryonale Organismus gelernt hat, die Zellen eines andern Embryos wie eigene Zellen zu behandeln, wird er im erwachsenem Zustand den Ge- weben des Spenders mit der gleichen Toleranz begegnen. Medawar (3-^1) u^d andere Autoren haben den Begriff der „inmunitaren Toleranz" auf Phánomene ausgedehnt, die sich nicht nur auf Zellen beziehen, sondern audi auf andere Antigene wie Proteine und Polisaccharide der Pneumokokken. Atopische und anaphylaktisché Phánomene, die auf einer Antigen-Antikórperreak- tion beruhen, besitzen in bezug auf die Antigenreaktion eine enge Beziehung zum Inmunitátsbild. Auf dem Erwahnten basiert vorliegende Arbeit; es solí nach Móglichkeit ein Tole- Tanzzustand gegenüber heterologen Proteinen bei Meerschweinchen erreicht werden, d. h. bei einer Spezies, die sich besonders leicht in einen anaphylaktischen Zustand versetzen laBt.

    Etiological agents of respiratory allergy in tropical countries of Central and South America

    In the tropical countries of Central and South America there grow only a few species of plants that produce allergenic pollen, and these are not widely distributed, with the exception of the grasses which otherwise have a feeble sensitizing power. The output of pollens from grasses is very limited in the high mountains because of the lack of flowering of such plants. The most important species correspond to the families Amaranthaceae, Chenopodiaceae, and Compositae.
