| Abstract: | The following research work sought to develop an optimal method of concentration and recovery of various commonly used medications, present in a water matrix, by using the solid phase extraction technique or by its acronym SPE (Solid Phase Extraction), which uses cartridges (columns with different supports that adsorb a wide variety of molecules according to their chemical structure). It consists of four main stages, conditioning of the cartridge, loading of the sample, washing of interferences, elution of the analytes of interest by means of a solvent. For the experimental design, three independent variables were taken into account (pH of the water sample, elution solvent and its pH), three types of cartridges were evaluated (anion exchange, cation exchange and reverse phase), each experiment was performed with solutions aqueous medications at known concentrations to calculate the recovery percentage. Once each test was completed, the concentration was measured by HPLC chromatography, and the recovery percentages were calculated from the results.
At the end of the research work, an SPE method was optimized for the recovery of the five drugs analyzed, starting with solutions of 20µg/L, four of them, Sulfamethoxazole, Ciprofloxacin, Diclofenac and Ibuprofen, using the Hypersep PEP cartridge (reverse phase) obtaining percentages of recovery of 111%, 86%, 88% and 80% respectively and Paracetamol using the Hypersep AX cartridge, with a recovery percentage of 98%. |
| Description: | En el siguiente trabajo de investigación se buscó desarrollar un método óptimo de concentración y recuperación de distintos medicamentos de uso común, presentes en una matriz de agua, mediante el uso de la técnica de extracción en fase sólida o por sus siglas SPE (en inglés Solid Phase Extraction), que utiliza cartuchos (columnas con distintos soportes que adsorben una gran variedad de moléculas de acuerdo a su estructura química). Consta de cuatro etapas principales, condicionamiento del cartucho, carga de la muestra, lavado de interferencias, elución de los analitos de interés por medio de un solvente. Para el diseño experimental se tomó en cuenta tres variables independientes (pH de la muestra de agua, solvente de elución y su pH), se valoró tres tipos de cartuchos (intercambio aniónico, intercambio catiónico y fase reversa), cada experimento se realizó con soluciones acuosas de los medicamentos a concentraciones conocidas para poder calcular el porcentaje de recuperación. Una vez concluido cada ensayo se procedió a medir la concentración por cromatografía HPLC, y a partir de los resultados se calculó los porcentajes de recuperación.
Al final del trabajo de investigación se optimizó un método SPE para la recuperación de los cinco fármacos analizados, partiendo de soluciones de 20µg/L, cuatro de ellos, Sulfametoxazol, Ciprofloxacina, Diclofenaco e Ibuprofeno, mediante el cartucho Hypersep PEP (fase reversa) obteniéndose porcentajes de recuperación del 111%, 86%, 88% y 80% respectivamente y el Paracetamol mediante el cartucho Hypersep AX, con un porcentaje de recuperación del 98%. |
