    Estandarización de la metodología de cromatografía de columna para el aislamiento de metabolitos activos a partir de extractos vegetales

    The present research aimed to standardize the methodology of column chromatography, for which a general phytochemical march of the species Desmodium adscendens was performed by thin layer chromatography (TLC), obtaining positive results for the active metabolites flavonoids and terpenes. The fractionation of column chromatography focused on obtaining representative amounts of flavonoids due to their known anti-inflammatory and antioxidant properties, using the following variables: mobile phase selection and column loading. The free dry methanolic extracts of the chlorophyll excess from the aforementioned species were eluted in the columns with mobile phases of polar and nonpolar characteristics and with a load of 2.5% and 5%, each fraction collected was evaluated in a TLC analysis Obtaining as results that the flavonoids possess greater affinity for the polar mobile phase and that the column load with better efficiency and resolution is 2.5%. Finally, the qualitatively identical fractions corresponding to the flavonoids were united to calculate their final yield and to perform the analysis of their antioxidant activity, giving positivity to the same with DPPH. Column chromatography turned out to be an unreliable technique probably due to the influence of environmental factors such as the inter-day and intra-day temperature, so it was not possible to standardize the method.
