In this study, 40 clinically healthy male dogs were used as experimental units, assessed by the ASA classification system (American Society of Anesthesiologists); divided into two groups: puppies from 6 to 12 months weighing 7 to 15kg and adults over 12 months to 60 months weighing from 11 to 25 kg, using a pre-scrotal approach, under the same anesthetic protocol, were randomly assigned to the ligature techniques: using Vicryl (T1) and making square knots (T2). Three blood samples were taken and analyzed, one pre-surgical, at 24 hours and 8 days after surgery. This investigation was descriptive and experimental. The variables analyzed were: surgery time, inflammation (blood count values, leukogram), pain assessment (Glasgow scale modified by Otero) and physiological constants (HR, RR, T°, SpO2) resulting in: no significant time variables of surgery, physiological parameters and pain assessment. There was a significant difference (P<0,05) in platelets of adult dogs at 8 days in T1 307,2±143,13 and T2 294,8±139,34; in band neutrophils T1 7,5±23,49 and T2 1,3±2,40; Leukogram in T1 and age 1 for band neutrophils between the pre-surgical 0,32±0,093 and 8 days 7,04±6,462, and between 24 hours 0,94± 0,372 and 8 days 7,04±6,462. Concluding that with the surgical technique T1 (Vicryl) there was greater inflammation in adult dogs, observing an increase in band neutrophils; and in the assessment of pain, patients submitted to T2 presented a better behavior in relation to T1
