Ajuste del tiempo de inseminación con semen sexado en vacas holstein en producción superovuladas
(2018) Argudo Garzón, Daniel Ernesto; Soria Parra, Carlos Alonso; Durán López, Juana Catalina; Bravo Mosquera, Sandra Elizabeth; Alvarado Molina, Juan Pedro; Alberio, Ricardo Horacio
The embryo production results after use of sex-sorted sperm to inseminate superovulated cows (SOV) have not been very encouraging so far, due to the low quantity of sperms used as well as their short half- life. In a previous study conducted in Brazil in Holstein cows, it was shown that using the protocol of SOV P36/Lh60 and inseminating with sexsorted sperm at 18 and 30h (interval of 12hs between the two inseminations) after application of the inductor of ovulation it was obtained a similar quantity of transferable structures than inseminating at the 12 and 24h with non-sorted sperm. The aim of this investigation was to adjust the moment of the artificial insemination (AI) in superovulated and inseminated cows with sex-sorted sperm to achieve better synchrony with ovulations and by this way to increase the quantity of transferable embryos obtained. For this, we evaluated 2 moments of AI in which the interval between both were of 6hs. We used 30 lactating Holstein cows treated with the protocol P36/LH60 and, at insemination, were divided at random in three groups: 1-group AI18/30 (control group; n=10), the AI were carried out at the 18 and 30h after application of the inductor of the ovulation (GnRH); 2- group AI18/24 (n=10) the AI inseminations were made at 18 and 24 h after GnRH and 3- group AI24/30(n=10) the AI were made at 24 and 30h. In all cases, each insemination was doing with 2.1 x 106 sex-sorted sperm. The results between groups were no different in the quantity of transferable embryos (1,9±0,6;4,1±1,5 and 1,3±0,4respectively; p>0,05) nor in the proportion of transferable from total structures (38,8%; 60,1% y 34,1% respectively). In conclusion, the low number of cows in each group and the high variability of responses do not permit to conclude on significant differences in the alternatives evaluated. However, the numerical results lead to propose new studies to increase the number of animals treated and so, reevaluate these results.

Ajuste del tiempo de inseminación con semen sexado en vacas superovuladas
(2016) Argudo Garzón, Daniel Ernesto; Soria Parra, Carlos Alonso
The results of the combination of sex-sorted sperm and superovulation (SOV) have not been very encouraging so far, due to the low quantity of sperms as well as their short life. In a previous study conducted in Brazil in Holstein cows, it was shown that using the protocol of SOV P36/Lh60 and inseminating with sex-sorted sperm at the 18 and 30 hours of application of the inductor of the ovulation (interval of 12 hs between the two inseminations) it is obtained an equal quantity of transferable structures than inseminating at the 12 and 24 hours with non-sorted sperm. The aim of this investigation was to adjust the hours of the artificial insemination (AI) in superovulated and inseminated cows with sex-sorted sperm to achieve better synchrony with ovulations, so in this way to increase the quantity of transferable embryos. This adjustment evaluated 2 moments of AI in which the interval diminished to 6 hs. For this, 30 superovulated lactating Holstein cows were used with the protocol P36/LH60 and were divided in three groups in an aleatory form; in the group IA18/30 (control group; n=10) the inseminations were carried out at the 18 and 30 hs of application of the inductor of the ovulation (GnRh); in the group IA18/24(n=10) was inseminated at the 18 and 24 hs of the GnRH and the group IA24/30(n=10) was inseminated at the 24 and 30 hs (it was used 2.1 x 106 sex-sorted sperm/insemination). It was not found statistical difference in the three groups; however, the group IA18/24 showed numeric advantage over the group IA24/30 and the Control (4,1±1,5 vs 1,3±0,4 and 1,9±0,6 respectively) in the quantity of transferable embryos, concluding that this adjustment in the hours of AI with sexed semen in superovulated lactating Holstein cows can be used with similar results to the previous works

Determinación del porcentaje de preñez con protocolos IATF en vacas lecheras utilizando benzoato y cipionato de estradiol
(2014) Calva Calva, Jean Carlo; Cantos Torres, Edwin Patricio; Soria Parra, Carlos Alonso
The investigation was made in Tarqui, Cumbe and Victoria del Portete in order to determinate the pregnancy percentage of the protocols. A sample of 40 cows was taken for this investigation, which was divided in 20 animals for each treatment. The protocols were applied in the following way: Day 0, a Progestin intravaginal device (CIDR) was placed, 2 mg of estradiol benzoate (EB) IM, on day 8 the CIDR was removed, 150ug of PGF2 α or its synthetic analog and 400 UI of ECG was applied. Later the cows were divided in two groups. First group received 1mg of BE on day 9, while group number two received 1mg of estradiol cypionate (CPE) on day 8, finally the insemination was made from 56 to 60 hours after the CIDR removal. On this investigation a 55% average pregnancy percentage was obtained. T student test shows that there is no statistical difference between treatments and the ANOVA indicates that there is no difference between the CPE and the BE regarding corporal condition.

Efecto de progesterona inyectable (p4) aplicada en el día 3 post-inseminación sobre el cuerpo lúteo y la fertilidad en vacas Brahman y Brown Swiss en el trópico ecuatoriano
(2017) Macas Apolo, Freddy Fausto; Soria Parra, Carlos Alonso
This study evaluated the effect of applying exogenous progesterone on the corpus luteum three days after artificial insemination at fixed time (IATF) in Brahman and Brown Swiss cows, aged 4 to 6 years with condition Body weight between 2.5 and 3 on the scale of 1 to 5. This research was carried out on 29 cows divided into two farms located in the southern tropical zone of Ecuador; In a farm with 15 Brahman cows (n = 5 control group, n = 5 group75 mg, n = 5 group 100 mg) and in another 14 Brown Swiss cows (n = 5 control group, n = 4 group75 mg, n = 5 Group 100 mg). According to the chronogram that was established for this purpose, ultrasound was performed to determine the growth of the corpus luteum (CL) a day over another. It also proceeded to draw blood from the coccygeal vein in all cows for establish the plasma concentration of progesterone (P4) ng / ml. Statistical analysis to determine serum P4 levels and CL morphology was performed using the ANOVA test and to determine the pregnancy diagnosis was performed using the chi-square test, this diagnosis was made with an ultrasound at 60 days after insemination, giving As a result 40% of the Witness group, 44.4% 75 mg and 50% of the 100 mg group. However the treatments evaluated did not present significant differences between them (P > 5), therefore the different doses applied not interfere with development of CL therefore not significantly effect on pregnancy rate

Effect of estradiol benzoate on the size of follicle and corpus luteum of Holstein cows treated with D-cloprostenol (2015)
(2015) Soria Parra, Manuel Elías; Narvaez Teran, Jhonny Alfredo; Torres Inga, Carlos Santiago; Méndez Álvarez, María Silvana; Soria Parra, Carlos Alonso
The effect of two treatments, farms, and body shape on the size of the follicle and Corpus Luteum were evaluated. D-Cloprostenol (150 ug) was used in one of the treatments; in the other, D-Cloprostenol (150 ug) and Estradiol Ben-zoate (1 mg), were used. Two groups of 32 Holstein dairy cows each, from the Nero and Irquis farms in Cuenca, Ec-uador, were set up. Body condition (2.75-3.50) was considered. The calving number was 1-6; the farms, Irquis and Nero; and animal age, 3-10 years. Ultrasound scanning was used on days 0; 3 and 7, in order to measure the Luteum first, then the dominant ovarian follicle, and finally, the Corpus Luteum. Factor analysis of variance was performed. No significant differences were found for follicle size due to any factors; nor due to interaction. Concerning luteum, significant differences (P < 0.05) were observed for the treatments and for the farm. The values were 0.33 cm higher for the treatment with Benzoate, and 0.36 cm, at Irquis, in comparison to Nero. The treatment with Benzoate, along with better husbandry at Irquis played a critical role in the appearance and evolution of the luteum.
