Item Optimización de la producción del antígeno recombinante VP2 del Protoparvovirus carnivoro 1 en Escherichia coli(Universidad de Cuenca, 2025-01-09) Guasco Escobar, Luis Marcelo; Vallecillo Maza, Antonio JavierCarnivorous protoparvovirus 1 is the viral species currently assigned to the causal agent of canine parvovirosis, a common disease in puppies and for which several clinical forms have been described, including hemorrhagic gastroenteritis, myocarditis, cerebellar hypoplasia, cerebelitis or encephalitis. The ability to immediately detect the disease in puppies is important for the prevention and control of the disease. Biotechnological tools allow the development of assays for the detection and control of the disease, so in the present study the production of virion-like particles was implemented, using a bacterial expression model; in which the culture conditions were identified, and then the purification and formation of viral pseudoparticles were optimized. A hemagglutination and hemagglutination inhibition assay was then applied to demonstrate the formation of virion-like particles. Finally, the hemagglutination inhibition assay allowed a comparison of the presence of antibodies in patients immunized with viral antigen versus patients who recovered from the disease; demonstrating the usefulness of virion-like particles to demonstrate the presence of antibodies, which, in these two contrasted groups, no significant differences were found (p > 0.05). Therefore, for future research it will be important to study with the use of virion-like particles, among other variables, the immunological status of patients recovered from infections by existing viral variants that can infect canines.
