Item Evaluación de diferentes protocolos de maduración y fecundación in vitro de COC´S en bovinos(Universidad de Cuenca, 2022-09-20) Espinoza Siguencia, Luis Manuel; Calle Zárate, James Genaro; Ayala Guanga, Luis EduardoThe objective of this research was to evaluate the efficiency of three protocols for in vitro maturation and fertilization of bovine oocytes, using two media produced in the Biotechnology laboratory of the Faculty of Agricultural Sciences (in house 1 and 2), versus commercial media. Two experiments were carried out, the first one determined the efficiency of in vitro maturation and the second one the in vitro fertilization. The treatments corresponded to two media elaborated in the laboratory "in house 1 = T1 and in house 2 = T2" and a commercial medium "BO-IVMTM= T3", which was used as a control. The results determined the existence of statistical difference in the expansion of cumulus cells (p<0.05) in grade 2, being lower in T1 (23.9%) and T2 (18.4%) in relation to T3 (30.7%), and in grade 3, T1 (65.9%) and T2 (71.1%) their percentages were similar, but T3 presented a lower percentage 59% to the two previous treatments (P<0.05). In the evaluation of meiosis progression, in the germinal vesicle stage, no significant differences were found in the treatments, but in Metaphase II, T3 (82.60%), T2 (69.80%), the latter has similar values with in house 1 (75.60%). Finally, in the formation of pronuclei, after in vitro fertilization, statistical differences were evidenced between T3 (89.5%) and T1 (79.3%); moreover, between T1 and T2 (70.9%) difference was observed between the treatments (p<0.05). Therefore, it is concluded that the in vitro maturation medium in house 1 elaborated in the laboratory is as efficient as the commercial medium. However, T3 medium allows obtaining a higher percentage of pronuclei after 24 hours of IVF.
