    Efecto de la idebenona en la congelación y vitrificación de semen ovino

    Idebenone (IDB) is a potent antioxidant that has proven effective in mitigating oxidative stress during the cryopreservation of ram semen. This study evaluated the effect of IDB on conventional slow freezing (CS) and kinetic vitrification (VIT) of ovine spermatozoa. In Experiment 1, fourteen ejaculates from four rams were used to determine the optimal IDB concentration. A dose of 50 µM was identified as optimal (compared to 0, 10, 25, and 100 µM) based on improvements in total motility (TM) and beat cross frequency (BCF) of spermatozoa stored at 5 °C for up to 96 hours. In Experiment 2, twenty-seven ejaculates from the same rams were assigned to four treatments based on the cryopreservation method and the presence or absence of 50 µM IDB: CS+IDB50, CS-Control, VIT+IDB50, and VIT-Control. The CS+IDB50 treatment significantly improved (P < 0.05) TM, progressive motility, viability, and acrosomal integrity, and significantly reduced DNA fragmentation compared to CS-Control. In contrast, although the VIT+IDB50 treatment reduced DNA fragmentation (P < 0.05), it also caused an undesirable decrease (P < 0.05) in curvilinear, straight-line, and average path velocities, as well as in straightness, linearity, amplitude of lateral head displacement, and BCF compared to VIT-Control. In conclusion, the addition of IDB improved motility, viability, acrosomal integrity, and DNA integrity in conventionally frozen ram spermatozoa, whereas it negatively affected sperm kinematic parameters in vitrified samples.
