Efecto del resveratrol sobre la congelación y vitrificación de espermatozoides epididimarios de perro

Abstract

This research assessed the effect of Resveratrol (RES) on the conventional slow freezing (CS) and vitrification (V) of canine epididymal sperm on cryosurvival. Twenty sperm pooled samples from 60 epididymal from 30 orchiectomized dogs were used in this study. Each pool was diluted with TCG-YH (6% egg yolk), and then 4 treatments were conformed based on the type of cryopreservation and the addition of 100 µM RES: 1) CS-RES, 2) CS-Co[control], 3) V-RES, and 4) V-Co[control]. Freezing was performed by exposing 0.25 ml straws with sperm samples (with 5% glycerol) to static liquid nitrogen (LN2) vapors. Vitrification was done by directly dropping 30-μL drops of samples (with 250 mM sucrose) into LN2. The CS-RES treatment increased (P<0.05) post-thaw values of total motility, rapid curvilinear velocity (VCL), and the amplitude of lateral head displacement (ALH), and integrity of plasma and acrosome membranes, and reduced oxidative stress. However, the V-RES treatment did not induce any beneficial effect (P>0.05) after warming. Vitrification treatments increased (P<0.05) DNA fragmentation compared to freezing treatments. The CS-RES treatment increased (P<0.05) the dimensions of sperm head after freezing. In conclusion, RES had a beneficial and enhanced effect on motility, membrane protection, and exerted an antioxidant effect during and after the freezing of canine epididymal sperm; however, its effect was limited in vitrification.
