Abstract: | The taeniasis/cysticercosis complex is a worldwide zoonotic parasitic disease
caused by the tapeworm Taenia solium, T. saginata and T. asiatica. The species
that affect pigs and humans are T. solium and T. asiatica. The natural cycle of the
parasite includes the pig as an intermediate host and the human as the final host
and/or in some cases as an intermediate host due to fecal-oral contamination.
Ecuador is considered an endemic country to human taeniasis and hyper-endemic
to Neurocysticercosis. While in pigs, studies and prevalence figures do not report
on the evolution of the number of cases over time and estimates of the incidence
rate of cysticercosis are lacking, causing great economic losses and being a
serious health problem. The objective of the present study was to identify larval
stages of T. solium and T. asiatica in tissue samples from pigs (Sus scrofa ssp.
domestica) using the LAMP biomolecular technique; to generate a first information
on its use and viability. A total of 76 samples of cysts with lesions were collected in
different tissues of the pig suspected of larval stages of Taenia spp. with a greater
location in the liver (52), followed by masseter muscle (5,) shoulder butt (4), tongue
(4), biceps (3), heart (2), diaphragm (2), thigh (2), neck muscle (1) and intestine
(1). The viability of the samples (process control) was determined by a LAMP
assay for Sus scrofa spp. domestica with a set of primers for the COXII gene, while
a LAMP assay was used to identify the samples of T. solium and T. asiatica with a
set of primers for the COXI genes. As a result, it was observed that, of the total
samples collected, 36.84% were valid for process control, while for the LAMP test
for T. solium, 15.79% of positive samples were obtained; to finally have a total of
10.53% positive samples and also valid for process control, on the contrary, for T.
asiatica all the samples were negative. The LAMP technique is capable of
detecting larval stages positive to T. solium and valid for the control of the process,
such as the case of intestine and various liver tissue samples, which could easily
be confused with E. granulosus or T. hydatigena. However, it is evident that
several of the samples that coincide with lesions characteristic of cysticercosis,
were negative in the test for T. solium and not valid in the control of the process.
Being necessary to adapt the tests for the reactives used, improve the protocols in
the extraction method or even take into account the amount of porcine tissue that
covers the cysts and seek more evidence that may suggest whether or not the
viability of these may affect the way in which DNA is preserved. |
Description: | El complejo teniasis/cisticercosis es una enfermedad parasitaria zoonótica de
distribución mundial, causada por el cestodo Taenia solium, T. saginata y T.
asiatica. Las especies que afectan al cerdo y al humano son T. solium y T.
asiatica. El ciclo natural del parásito incluye al cerdo como hospedero
intermediario y al humano como hospedero definitivo y/o en algunos casos como
hospedador intermediario por contaminación fecal-oral. El Ecuador se considera
un país endémico a teniasis humana e hiper-endémico a la Neurocisticercosis.
Mientras que, en cerdos, los estudios y las cifras de prevalencia no informan sobre
la evolución del número de casos a lo largo del tiempo y faltan estimaciones de la
tasa de incidencia de cisticercosis, provocando grandes pérdidas económicas y
siendo un grave problema de salud. El objetivo del presente estudio fue identificar
estadios larvarios de T. solium y T. asiatica en muestras de tejidos de cerdos (Sus
scrofa ssp. domestica) mediante la técnica biomolecular de LAMP; para generar
una primera información sobre su uso y viabilidad. Se colectaron un total de 76
muestras de quistes con lesiones en diferentes tejidos del cerdo sugerentes a
estadios larvarios de Taenia spp. con una mayor ubicación en hígado (52),
seguido de músculo masetero (5,) espaldilla (4), lengua (4), bíceps (3), corazón
(2), diafragma (2), muslo (2), músculo del cuello (1) e intestino (1). La viabilidad de
las muestras (control del proceso) se determinó mediante un ensayo LAMP para
Sus scrofa spp domestica con un set de cebadores para el gen COXII, mientras
que para identificar las muestras de T. solium y T. asiática se utilizó un ensayo
LAMP con un set de cebadores para los genes COXI. Como resultados del total de
muestras colectadas, el 36,84% fueron válidas al control del proceso, mientras que
para el ensayo LAMP para T. solium se obtuvo un 15,79% de muestras positivas;
para finalmente tener un total de 10,53% muestras positivas y también válidas al
control del proceso, por el contrario, para T. asiática todas las muestras resultaron
negativas. LAMP es capaz de detectar estadios larvarios positivos a T. solium y
válidos al control del proceso, como el caso de las muestras de intestino y varios
de tejido hepático, los cuales fácilmente podían ser confundidos con E. granulosus
o T. hydatigena. Sin embargo, es evidente que varias de las muestras que
coinciden con lesiones características a estadios larvarios de tenia, resultaron
negativas al ensayo para T. solium y no válidas al control del proceso. Siendo
necesario adaptar los ensayos para los reactivos utilizados, mejorar los protocolos
en el método de extracción o incluso tener en cuenta la cantidad de tejido porcino
que cubre a los quistes y buscar mayor evidencia que pueda sugerir si la viabilidad
de estos puede afectar o no en la forma en cómo se logra preservar el ADN. |
