Abstract: | The canine parvovirus is a contagious disease caused by canine parvovirus type 2 (CPV-2). It especially affects puppies under six months and adult dogs without immune protection. The signology includes lethargy, anorexia, fever, vomiting and diarrhea; if the animal doesn’t get support treatment, the illness can be lethal. The
original strain CPV-2 has been completely replaced by three variants (CPV-2a, CPV-2b, CPV-2c), and this evolution has occurred in a relatively short time.
However, there is controversy regarding how virulent these strains are, as well as the level of protection of the current vaccines. The aim of this study was to identify the different strains of CPV-2 in dogs with parvovirosis compatible signs in the city of Cuenca. For this, 80 feces samples were collected, from which those that did
not generated a specific amplicon of dog (Process control) were discarded. Subsequently, the disease diagnostic PCR was performed and 25 positive samples were selected and sent to sequencing. From these samples, it was possible to obtain the genetic sequence of 21. The 47,61% belonged to strain CPV-2a, followed by strain CPV-2c with 38.1%, and finally the strain CPV-2b with 14,29%. According to what has been published, these strains suffer prevalence changes and can generate subvariants of itselfs over the time and in different geographic locations. |
Description: | La parvovirosis canina es una enfermedad infecto-transmisible causada por el Parvovirus canino tipo 2 (CPV-2). Afecta especialmente a cachorros menores de seis meses y a perros adultos sin protección inmunológica. La signología incluye letargia, anorexia, fiebre, emesis y diarrea; si el animal no recibe tratamiento de soporte, la enfermedad puede ser fatal. La cepa original CPV-2 ha sido reemplazada completamente por tres variantes (CPV-2a, CPV-2b, CPV-2c), y dicha evolución se ha dado en un relativamente corto periodo de tiempo. Por otro lado, existe controversia con respecto a cuan virulentas resultan estas cepas, así como el nivel de protección de las vacunas actuales. El objetivo de este estudio fue identificar las diferentes cepas de CPV-2 en perros con signología compatible a parvovirosis en la ciudad de Cuenca. Para ello se recolectó 80 muestras de
heces, de las cuales se descartaron las que no generaron un amplicón específico de perro (Control de proceso). Posteriormente se realizó el PCR de diagnóstico de la enfermedad y se seleccionaron 25 muestras positivas que fueron enviadas a secuenciación. De estas muestras, fue posible obtener la secuencia genética de 21. El 47,61% pertenecieron a la cepa CPV-2a, seguida por la cepa CPV-2c con un 38,1%, y por último la cepa CPV-2b con un 14,29%. De acuerdo a lo publicado estas cepas sufren cambios de prevalencia y pueden generar subvariantes de las mismas en el tiempo y en diferentes ubicaciones geográficas. |
