| Abstract: | The research was conducted in the biotechnology laboratory. The objective was to determine the effect of the semen sexing agent (HeiferPlus™) on the percentage of female embryos obtained in vitro production (PIV). Sperm quality was assessed, number of clivaled oocytes and blastocysts obtained and number of embryos female and male. Three times of co-incubation (semen + HeiferPlus™) and one control were analyzed, using a completely randomized design, the results were evaluated in SPSS® version 25. The qualitative variables were compared using Chi-square test, for the quantitative ones performed an ANOVA and to compare means the Tukey test at 5%. Treatments T2 and T3 differ from T1 and control in terms of Vitality and Progressive Individual Motility; Membrane Integrity decreased in T1, T2 and T3 versus control; The Abnormalities and Integrity of the Acrosomal Membrane showed no difference. The cleavage of T3 decreased with respect to T1 and T2, which have lower values than the control. The number of embryos was similar T2, T3 and control, decreasing in T1. The ratio of female and male embryos do not differ statistically, although an increase of 20.5% and 19.4% was found in favor of the T1 and T2 females compared to the control. In conclusion HeiferPlus™ partially affects the seminal quality, resulting in a lower number of cleaved oocytes; however, it does not affect the number of embryos obtained; however, there is a significant non-statistical increase in female embryos in PIV when using the sexing agent |
| Description: | La investigación se realizó en el laboratorio de biotecnología. El objetivo fue determinar el efecto del agente sexador de semen (HeiferPlus™) sobre el porcentaje de embriones hembras obtenidos en producción in vitro (PIV). Se valoró calidad espermática, cantidad de ovocitos clivados y blastocistos obtenidos y número de embriones hembras y machos. Se analizó tres tiempos de co-incubación (semen + HeiferPlus™) y un control, utilizando un diseño completamente al azar, los resultados se evaluaron en SPSS® versión 25. Se comparó las variables cualitativas utilizando prueba de Chi cuadrado, para las cuantitativas se realizó un ANOVA y para comparar medias la prueba de Tukey al 5%. Los tratamientos T2 y T3 difieren de T1 y control en cuanto a Vitalidad y Motilidad Individual Progresiva; la Integridad de Membrana disminuyó en T1, T2 y T3 frente al control; las Anormalidades e Integridad de la Membrana Acrosomal no presentaron diferencia. El clivaje de T3 disminuyó respecto a T1 y T2, los cuales presentan valores menores que el control. El número de embriones fue similar T2, T3 y control, disminuyendo en T1. La relación de embriones hembras y machos no difieren estadísticamente aunque se encontró un incremento del 20,5% y 19,4% a favor de las hembras T1 y T2 frente al control. En conclusión HeiferPlus™ afecta parcialmente la calidad seminal, resultando un menor número de ovocitos clivados; sin embargo, no afecta el número de embriones obtenidos; empero, existe un incremento significativo no estadístico de embriones hembra en PIV al usar el agente sexador |
