Abstract:
The present research is a non-experimental cross-sectional study with case and control design. The aim of the study was to genotypic and phenotypically association of the SNP 2548G>A of the LEP gene with the presence of type 2 diabetes mellitus in outpatients of the DONUM Foundation. A total of 116 patients were enrolled and divided into two equal groups (n = 58), according to the previous diagnosis of DM2, the groups were paired by age, gender and body mass index. The history of cardiovascular, cerebrovascular, renal diseases, immunodeficiency diseases; and gestation status were exclusion criteria. Data collection was done through the direct interview using a survey; EDTA tubes were used to collect blood samples; later, it was performed the DNA extraction, and the genetic material was amplified using the real time PCR technique.
The Statistical analysis was performed with 95% confidence interval and p <0.05. The presence of the A allele and the AA genotype was found with higher frequency (16%) in the DM2 group, compared with the controls (12%); however, no statistically significant association was found between the presence of the allelic variant and the development of Type 2 Diabetes Mellitus.

El presente trabajo es un estudio no experimental de tipo transversal con diseño de casos y controles. El objetivo fue asociar genotípica y fenotípicamente el polimorfismo 2548G>A del gen LEP con la presencia de diabetes mellitus tipo 2 en pacientes ambulatorios de la fundación DONUM. Se trabajó con un total de 116 pacientes divididos en dos grupos de igual número (n=58), de acuerdo al diagnóstico previo de DM2; las variables edad, género e índice de masa corporal se aplicaron como criterios de pareamiento. Los antecedentes de enfermedades cardiovasculares, cerebrovasculares, renales, enfermedades de inmunodeficiencias y el estado de gestación fueron criterios de exclusión. La recolección de datos se realizó mediante entrevista directa con aplicación de encuesta; para la toma de muestras de sangre se emplearon tubos con EDTA, posterior se realizó la extracción del ADN, y se amplifico el material genético mediante la técnica de PCR real time.
Para el análisis estadístico se trabajó con un intervalo de confianza del 95% y p<0.05. La presencia del alelo A y el genotipo AA se encontró con mayor frecuencia (16%) en el grupo con DM2, comparado con los controles (12%); sin embargo, no se encontró asociación estadísticamente significativa entre la presencia de la variante alélica con el desarrollo de Diabetes Mellitus Tipo 2.
