The present research aimed to standardize the methodology of column chromatography, for which a general phytochemical march of the species Desmodium adscendens was performed by thin layer chromatography (TLC), obtaining positive results for the active metabolites flavonoids and terpenes. The fractionation of column chromatography focused on obtaining representative amounts of flavonoids due to their known anti-inflammatory and antioxidant properties, using the following variables: mobile phase selection and column loading.
The free dry methanolic extracts of the chlorophyll excess from the aforementioned species were eluted in the columns with mobile phases of polar and nonpolar characteristics and with a load of 2.5% and 5%, each fraction collected was evaluated in a TLC analysis Obtaining as results that the flavonoids possess greater affinity for the polar mobile phase and that the column load with better efficiency and resolution is 2.5%. Finally, the qualitatively identical fractions corresponding to the flavonoids were united to calculate their final yield and to perform the analysis of their antioxidant activity, giving positivity to the same with DPPH.
Column chromatography turned out to be an unreliable technique probably due to the influence of environmental factors such as the inter-day and intra-day temperature, so it was not possible to standardize the method.

El presente trabajo de investigación pretendió estandarizar la metodología de cromatografía de columna, para lo cual se realizó una marcha fitoquímica general de la especie Desmodium adscendens mediante cromatografía en capa fina (TLC), obteniéndose resultados positivos para los metabolitos activos flavonoides y terpenos. El fraccionamiento de cromatografía de columna se enfocó en la obtención de cantidades representativas de flavonoides debido a sus propiedades antiinflamatorias y antioxidantes conocidas, usando las siguientes variables: selección de la fase móvil y la carga de columna.
Los extractos metanólicos secos libres del exceso de clorofila de la especie antes mencionada, fueron eluidos en las columnas con fases móviles de características polares y apolares y con una carga de 2,5% y 5%, cada fracción colectada fue evaluada en un análisis TLC obteniéndose como resultados que los flavonoides poseen mayor afinidad por la fase móvil polar y que la carga de columna con mejor eficacia y resolución es la de 2,5%. Finalmente, las fracciones cualitativamente idénticas correspondientes a los flavonoides fueron unidas para calcular su rendimiento final y para realizar el análisis de su actividad antioxidante, dando positividad a la misma con DPPH.
La cromatografía de columna resultó ser una técnica poco confiable probablemente por la influencia de factores ambientales como la temperatura inter-día e intra-día por lo que no fue posible la estandarización de la metódica.
