dc.description | La investigación evaluó el impacto del Resveratrol (RES) en la criosupervivencia de espermatozoides equinos, utilizando congelación convencional (C) y vitrificación cinética (V). Se recolectaron 16 muestras de semen de 4 caballos españoles y se dividieron en cuatro grupos: C-RES, C-Co, V-RES y V-Co, según el método de criopreservación (C o V) y la presencia de RES (10 o 0 μM). Durante la congelación, se utilizaron pajuelas de 0,5 mL con dimetilformamida al 5%, mientras que para la vitrificación se empleó 100 mM de trehalosa y sumergiendo las muestras en gotas de 30 μL en NL2. Los resultados revelaron que los tratamientos con RES, especialmente en C-RES, mejoraron la motilidad total y progresiva en comparación con V-RES y V-Co. Además, la velocidad curvilínea (VCL) fue más alta en C- RES y C-Co que en V-RES. Por otro lado, los valores de linealidad (LIN) y oscilación lateral (WOB) fueron más elevados en los tratamientos de V-RES y V-Co. Se observaron mayores niveles de integridad plasmática y acrosomal en C-RES, y menor fragmentación de ADN en estos grupos. También se detectó un efecto antioxidante del RES, reduciendo el estrés oxidativo en C-RES en comparación con C-Co. En conclusión, el RES mejoró la criosupervivencia de los espermatozoides equinos, evidenciándose en la motilidad y la integridad celular, y reduciendo la fragmentación del ADN y el estrés oxidativo. dc.description.abstract | The research assessed the impact of Resveratrol (RES) on the cryosurvival of equine sperm using conventional freezing (C) and kinematic vitrification (V). Sixteen semen samples from 4 Spanish horses were collected and divided into four groups: C-RES, C-Co, V-RES, and V-Co, based on the cryopreservation method (C or V) and RES presence (10 or 0 μM). During freezing, 0.5 mL straws with 5% dimethylformamide were used, while vitrification employed 100 mM trehalose and submersion in 30 μL drops in LN2. Results showed that RES treatments, especially C-RES, enhanced total and progressive motility compared to V- RES and V-Co. Additionally, curvilinear velocity (VCL) was higher in C-RES and C-Co than in V-RES. Conversely, linearity (LIN) and lateral oscillation (WOB) values were higher in V- RES and V-Co treatments. Increased levels of plasma and acrosomal integrity were observed in C-RES, with lower DNA fragmentation in these groups. An antioxidant effect of RES was also detected, reducing oxidative stress in C-RES compared to C-Co. In conclusion, RES improved equine sperm cryosurvival, as evidenced by enhanced motility and cellular integrity, along with reduced DNA fragmentation and oxidative stress.
