dc.description | La bioautografía directa es un método rápido y sencillo que presenta como ventaja la detección de la actividad antimicrobiana sobre placas cromatográficas, permitiéndonos así observar de una manera directa las fracciones que presentan dicha actividad. El presente trabajo consistió en la estandarización del método de bioautografía directa empleando extractos vegetales tanto polares como apolares con actividad antimicrobiana frente a Escherichia coli y Staphylococcus aureus. Además, para la cromatografía de capa fina (TLC) se utilizaron eluyentes acordes a la polaridad de los extractos vegetales, los cuales a su vez también fueron compatibles con los antibióticos gentamicina y ampicilina, usados como controles. Mediante esta técnica se pudo observar el crecimiento de S. aureus en las placas de TLC, sin embargo, tras varios ensayos no se logró visualizar a E. coli, por lo cual, se decidió implementar el método de bioautografía por inmersión o recubrimiento obteniendo como resultado el crecimiento de ambas bacterias y la visualización de las zonas de inhibición para los antibióticos y ciertos extractos mediante el empleo de reveladores como el MTT y la resazurina. No obstante, debido a la inestabilidad que presenta este método por todas las variables que deben ser controladas, no se pudo cumplir con el objetivo de estandarizar la bioautografía como herramienta de evaluación del potencial antibacteriano de extractos naturales. | en_US |
dc.description.abstract | Direct bioautography is a fast and simple method that has the advantage of detecting antimicrobial activity on chromatographic plates, allowing us to directly observe the fractions showing such activity. The present work consisted of the standardization of the direct bioautography method, by using both, polar and nonpolar plant extracts, with antimicrobial activity against Escherichia coli and Staphylococcus aureus. In addition, for thin layer chromatography (TLC), eluents were used according to the polarity of the plant extracts, which in turn were also compatible with the antibiotics gentamicin and ampicillin, used as positive controls. By means of this technique, it was possible to observe the growth of S. aureus in the TLC plates, however, after several tests, it was not possible to visualize the E. coli growth, therefore, it was decided to implement the bioautography method by immersion or coating, obtaining as a result the growth of both, bacteria and the visualization of the inhibition zones for the antibiotics and certain extracts by means of the use of MTT and resazurin. However, due to the instability of this method because of the different variables that must be controlled, the objective of standardizing bioautography as a tool for evaluating the antibacterial potential of natural extracts was not achieved. | en_US |
