dc.description | Una de las estrategias que se utilizan a nivel mundial para la multiplicación y conservación del material de alto valor genético o en peligro de extinción es la producción de embriones in vitro. El objetivo fue evaluar un protocolo de maduración in vitro de complejos cúmulo-ovocitos (COC’s) de cobayas (Cavia porcellus). Se obtuvieron 760 COC’s correspondientes a 52 ovarios, mediante la técnica de Slicing. Los COC´s fueron seleccionados de acuerdo a la metodología descrita por Hawk y Wall (1994) y modificada por Ayala et al. (2019) para COC´s de bovinos (tipo A; B; C). Una vez obtenidos los COC´s de cada tipo, el 20% fue utilizado para morfometría pre-maduración, otro 20% se utilizó como testigo y paso directamente al proceso de maduración in vitro (MIV) por 24 h. El restante 60% se aplicó la prueba del Azul Brillante de Cresilo (BCB) y se determinó el crecimiento (BCB+) y aquellos que aún estaban en crecimiento (BCB-), luego cada tipo y sus subgrupos (BCB+ y BCB-) fueron madurados in vitro durante 24 h. Finalmente se estableció el porcentaje de maduración y morfometría postmaduración. Los ovocitos recuperados fueron clasificados morfológicamente en un 35,8% de COC’s de tipo A, un 30,9% B y el 33,3% de tipo C. Se determinó que el 71,6%, 60%, 63,5% de COC´s tipo A, B y C respectivamente reaccionaron a la prueba BCB. Al realizar la morfometría se estableció que los COC´s de Tipo A y B (aptos), fueron más grandes que los COC´s no aptos (tipo C), con diferencia estadística. Los COC´s del grupo testigo de tipo A, maduraron en un 40,4%, los tipos B 31,7% y tipo C un 18,2%. Sin embargo, el porcentaje de maduración nuclear se vio afectado cuando los COC´s de los tipos A; B y C, fueron sometidos a la prueba del BCB y posteriormente colocados a madurar por 24 horas. En conclusión: el medio de maduración usado permitió que los ovocitos llegaran a metafase II; sin embargo, estos porcentajes están muy por debajo de lo esperado. | en_US |
dc.description.abstract | One of the strategies used worldwide for the multiplication and conservation of material of high genetic value or in danger of extinction is the production of in vitro embryos. The objective was to evaluate an in vitro maturation protocol for cumulus-oocyte complexes (COC´s) from guinea pigs (Cavia porcellus). 760 COC´s corresponding to 52 ovaries were obtained using the Slicing technique. The COCs were selected according to the methodology described by Hawk and Wall (1994) and modified by Ayala et al. (2019) for bovine COC´s (type A; B; C). Once the COC´s of each type were obtained, 20% was used for pre-maturation morphometry, another 20% was used as a control and went directly to the in vitro maturation process (IVM) for 24 h. The remaining 60% was applied the Brilliant Cresyl Blue (BCB) test and growth (BCB+) and those that were still growing (BCB-) were determined, then each type and its subgroups (BCB+ and BCB-) were matured. in vitro for 24 hours. Finally, the percentage of maturation and post-maturation morphometry were established. The recovered oocytes were morphologically classified as 35.8% type A COC´s, 30.9% B and 33.3% type C. It was determined that 71.6%, 60%, 63.5% of COC´s type A, B and C respectively reacted to the BCB test. When performing the morphometry, it was established that the COC´s of Type A and B (suitable) were larger than the unsuitable COC´s (type C), with a statistical difference. The COC´s of the control group of type A, matured in 40.4%, types B 31.7% and type C 18.2%. However, the percentage of nuclear maturation was affected when COC´s of types A; B and C, were subjected to the BCB test and subsequently matured for 24 hours. In conclusion: the maturation medium used allowed the oocytes to reach metaphase II; however, these percentages are much lower than expected. | en_US |
