dc.description | La bacteria Escherichia coli (E. coli) puede llegar a generar significativas pérdidas económicas en el sector agropecuario. Esta bacteria cuenta con un alto riesgo de contaminación biológica, resistencia a tratamientos antimicrobianos y alta zoonosis, sin embargo, hasta la fecha, los métodos de diagnóstico revelan únicamente el genotipo de la bacteria, pero no existen reportes en la zona sobre la identificación de los genes portadores de factores de virulencia (GFV). Por lo expuesto, el objetivo de esta investigación fue identificar GFV y la diversidad genotípica de cepas de E. coli seleccionadas por su fenotipo. Para conseguir el objetivo se trabajó con 30 cultivos positivos a la bacteria, aislados de pollos con signos clínicos criados en la provincia del Azuay. Las muestras fueron purificadas y posteriormente se realizó la tinción de Gram para cada cultivo. Mediante la técnica de PCR se detectó la presencia del gen uspA (específico de E. coli), también la presencia de GFV como son: adhesinas (fimC), exotoxinas (cvaA) y sistemas de captación-transporte de Hierro (iucD; chuA; fyuA). Finalmente, para revelar la diversidad genética se realizó la técnica (GTG)5. Se encontró la presencia de 25/30 (75%) cultivos que presentaron el gen uspA, permitiendo identificar E. coli, de ellos, la frecuencia de GFV positivos fue: chuA 12/25 (48%), cvaA 5/25 (20%), fimC 21/25 (84%), fyuA 9/25 (36%) y iucD 14/25 (56%). La diversidad genética por (GTG)5 manifestó la presencia de 2 grupos que abarcan los 30 cultivos bacterianos de este estudio. Con estos resultados podemos concluir que existen cepas de E. coli positivas al gen uspA, las mismas que contienen GFV que revelan una importante diversidad genética, de los pollos infectados con Colibacillosis de nuestro estudio. | en_US |
dc.description.abstract | Escherichia coli (E. coli) can cause significant economic loses in agricultural and animal industries. In spite of the high risks that this bacterium poses to food safety, environmental health and human health, as of today, most diagnostic methods focus only in the genotypic characterization of this bacterium’s isolates without addressing whether these isolates contain virulence factor genes (VFG). The objective of this work was to identify VFGs in isolates from commercial farms and to describe the genetic diversity of these isolates. Thirty isolates from chickens showing clinical signs of colibacillosis from commercial poultry farms in the province of Azuay (Ecuador) were used in the study. The isolates were purified and confirmed using gram-staining and PCR-amplification of the E. coli-specific genetic marker uspA. VFGs presence in the isolates DNA were screened using selected markers as follows: adhesins (fimC), exotoxins (cvaA), and iron uptake and transport systems (iucD, chuA and fyuA). The genetic diversity of the isolates was described using the (GTG)5 technique. About 75% percent (25/30) of the isolates reported the uspA marker confirming E. coli identity. Forty-eight percent (12/25) of the isolates reported the presence of the chuA marker, 20% (5/25) reported the cvaA marker, and 84% (21/25), 36% (9/25) and 56% (14/25) of the isolates reported the fimC, fyuA, and iucD markers, respectively. (GTG)5 analysis identified segregation of the bacterial isolates in two groups. These results reveal that most of the isolates contained VFGs and an large genetic diversity among the isolates. | en_US |
