Abstract: | The objective was to evaluate the Brilliant Cresyl Blue (BCB) in the selection of oocytes for the production of embryos in vitro (PIV). Three treatments were evaluated. G1 = Witness without BCB exposure; G2 = staining positive oocytes (BCB +) and G3 = staining negative oocytes (BCB-). Morphometry, maturation (IVM), meiotic configuration, cleavage and embryos were determined. 2,340 cumulus oocyte complexes (COCs) were obtained, classified as suitable and not suitable for IVP. The 2/3 parts were exposed for 90 min. To BCB and the rest incubated for 90 min in H-SOF. 25% of COCs from each group were taken to assess the morphometry. The remaining 75% was placed in maturing medium for 24h. After IVM, 25% of COCs were used for morphometry and meiotic progression. The remaining 50% was in vitro fertilized (IVF) and cultivated. Cleavage was evaluated 48h after IVF and embryos on day 8. 62.3% of G2 oocytes were BCB + and had a larger diameter before (135, 9 μm) and after (134, 8 μm) of IVM. (P <0.05) vs group 3 BCB- (128, 1 and 127, 6 μm) and the Witness (129, 0 and 128, 2 μm). Maturation and meiotic configuration were not different between groups (P> 0.05). Cleavage was higher (P <0.05) in the Control (43.4%) than the BCB + group (33.5%). Finally, the global percentage of embryos was higher in BCB + (52.6%) than the Controlling (32.6%) and BCB- (33.5%). It is concluded that the BCB allowed to select oocytes with a larger diameter, although it did not allow to increase the percentage of maturation and cleavage, it contributed to improving the overall percentage of embryos obtained in vitro. |
Description: | El objetivo fue evaluar el Azul Brillante de Cresilo (BCB) en la selección de ovocitos para la producción de embriones in vitro (PIV). Se valoraron tres tratamientos. G1= Testigo sin exposición al BCB; G2= ovocitos positivos a la tinción (BCB+) y G3= ovocitos negativos a la tinción (BCB-). Se determinó morfometría, maduración (MIV), configuración meiótica, clivaje y embriones. Se obtuvieron 2.340 complejos cúmulus ovocitos (COCs), clasificados en aptos y no aptos para la PIV. Las 2/3 partes fueron expuestos por 90 min., al BCB y los restantes incubados por 90 min en H-SOF. El 25% de COCs de cada grupo fue tomado para valorar la morfometría. El 75% restante se colocó en medio de maduración por 24h. Luego de la MIV un 25% de COCs se utilizó para la morfometría y la progresión meiótica. El 50% restante fue fertilizado in vitro (FIV) y cultivado. El clivaje se evaluó 48h después de la FIV y los embriones al día 8. El 62, 3% de ovocitos de G2 fueron BCB+ y presentaron un diámetro mayor antes (135, 9 μm) y después (134, 8 μm) de la MIV (P< 0, 05) vs el grupo 3 BCB-(128, 1 y 127, 6 μm) y el Testigo (129, 0 y 128, 2 μm). La maduración y la configuración meiótica no fueron diferentes entre grupos (P> 0, 05). El clivaje fue mayor (P< 0, 05) en el Testigo (43, 4%) que el grupo BCB+(33, 5%). Finalmente, el porcentaje global de embriones fue mayor en BCB+(52, 6%) que el Testigo (32, 6%) y BCB-(33, 5%). Se concluye que el BCB permitió seleccionar ovocitos con mayor diámetro, a pesar de que no permitió aumentar el porcentaje de maduración y clivaje, contribuyó a mejorar el porcentaje global de embriones obtenidos in vitro. |
