dc.description | El sistema de cultivo agroecológico promete el desarrollo de micro y macro organismos en el suelo como consecuencia una mayor fertilidad. Por el contrario, el sistema convencional suele ser criticado, se piensa que reduce la fertilidad del suelo como consecuencia de la aplicación permanente de productos sintéticos, lo cual tiene efectos en la diversidad y cantidad de la diversidad. En el país, no existen estudios comparativos entre estos dos sistemas de producción a nivel de comunidades microbianas. Las técnicas basadas en el aislamiento y cultivo de microorganismos limitan el conocimiento de estas comunidades ya que solo un grupo muy limitado de microorganismos pueden ser cultivados. Las técnicas actuales se basan en análisis moleculares y bioinformáticos. En este trabajo estudiamos las comunidades microbianas del suelo rizosférico de trigo cultivado de forma agroecológica y convencional, usando ADN del suelo. Para lo cual se tomaron 10 pares de muestras distribuidas en las provincias de Azuay y Cañar, cada par estuvo compuesto por una muestra de cada sistema de cultivo y por pares fueron colectadas en zonas similares, el historial del cultivo se evidenció a través de encuestas a los propietarios. El ADN metagenómico fue extraído de cada muestra y usado para amplificar la región ITS y 16S de la comunidad de hongos y bacterias respectivamente. El ADN amplificado fue sometido a DGGE y a partir del perfil de bandas generadas se analizó la diversidad de las comunidades usando los índices de Riqueza, Shannon y Equitatividad. No se encontraron diferencias significativas entre la diversidad de hongos ni bacterias al comparar ambos sistemas de cultivo. | es_ES |
dc.description.abstract | The system of agroecological cultivation promises the development of micro and macro organisms in the soil and consequently a greater fertility. On the contrary, the conventional system tends to be criticized because it is thought that it reduces the fertility of the soil as a result of the permanent application of synthetic products. In the country, there are no comparative studies between these two systems of production at the level of microbial communities. Techniques based on the isolation and cultivation of microorganisms limit the knowledge of these communities since only a very limited group of microorganisms can be cultivated. Current techniques are based on molecular and bioinformatic analysis. In this work we study the microbial communities of wheat rhizospheric soil cultivated in an agroecological and conventional way, using soil DNA. In total, 10 pairs of samples were collected in the provinces of Azuay and Cañar, each pair was composed of one sample of each culture system and in pairs were collected in similar areas, the history of the crop was evidenced through surveys the owners. The metagenomic DNA was extracted from each sample and used to amplify the ITS and 16S region of the community of fungi and bacteria respectively. The amplified DNA was submitted to DGGE and from the profile of generated bands the diversity of the communities was analyzed using the Wealth, Shannon and Equitativity indices. No significant differences were found between fungal and bacterial diversity when comparing both culture systems. | es_ES |
